Instructors: Jeffrey S. Rohrer, Ph. D. and Michael G. Hvizd Course Description: This one and a half day course will review the principles and practice of HPAE-PAD. The major areas of focus will be HPAE-PAD methods for monosaccharide, oligo- and polysaccharide, sialic acid, and sugar alcohol determinations. For each area of focus the participants will be instructed on the principals of the HPAE separation, column choice, and sample preparation for the most common applications. Common applications include the analysis of monosaccharides from glycoproteins, the analysis of monosaccharides from plants, the analysis of oligosaccharides from glycoproteins, the analysis of oligo- and polysaccharides from plants, the analysis of sialic acids from glycoproteins, and the analysis of carbohydrates in fermentation broths. This course will also include a review of the principals of pulsed amperometric detection for carbohydrates, instruction on using the eluent generation for monosaccharide analysis, and a brief review of the basic carbohydrate chemistry. Participants will be instructed on the practice of HPAE-PAD and the development of rugged HPAE-PAD methods. This includes instruction on eluent preparation, electrochemical cell maintenance, column care and maintenance, and chromatography system maintenance specific to HPAE-PAD. Participants will observe, and may assist in, eluent preparation and electrochemical cell assembly and installation to run to HPAE-PAD systems. These systems will run HPAE-PAD analysis of monosaccharides and oligosaccharides. All work will be done on ICS-3000 systems and participants should be experienced HPAE-PAD practitioners or have experience using an ICS-3000 system.
