Dionex Application Note 361: Extraction of Total Fat from Food Samples after Acid Hydrolysis using Accelerated Solvent Extraction (ASE) with GC/MS Analysis

March 2008, Sunnyvale, CA—Dionex is pleased to present a newly developed method for precise, accurate determination of total fats in food samples following acid hydrolysis: Application Note 361: Extraction Total Fat from Food Samples after Acid Hydrolysis Using Accelerated Solvent Extraction (ASE) with GC/MS Analysis . Determination of the total fat contents of food products continues to grow in importance because it is part measure of the total nutritive value of a food sample, and because it is required by law to be listed on prepared and packaged foods. The samples used in this study represent a cross-section of prepared packaged foods, including mayonnaise, corn chips, cheese, shortbread, and beef bologna.

Food samples can be problematic as extraction matrices, containing complex mixtures of ingredients, including both saturated and unsaturated fats. In order to obtain complete extraction of lipid compounds, samples must undergo acid hydrolysis. Hydrolyzed samples may cause corrosion of metal-based parts that come into contact with the acidic sample during extraction. The newly developed ASE systems from Dionex feature a pH-hardened flow path and extraction cells made of Dionium™, which is resistant to both moderately acidic and basic conditions, enabling an expanded range of extractions applications.

Accelerated Solvent Extraction is a method for using solvents at elevated temperatures and pressures to extract organic compounds for solid or semisolid materials. ASE® provides significant savings in labor and solvent used over traditional sample extraction methods such as Soxhlet, sonication, and shaking.
