Dionex offers several columns for the anion-exchange separation of amino acids. The new AAA-Direct TM Amino Acid System, which incorporates the AminoPac® PA10, revolutionizes the determination of amino acids because amino acids are detected directly, with high sensitivity, by
integrated pulsed amperometric detection (IPAD). This technique is recommended for the separation of free amino acids, for
example, in fermentation broths, protein characterization studies, or food analysis. The AminoPac PA1 offers the customer
a means to separate amino acids by anion-exchange separation and detect them using derivatization techniques such as OPA. For
the analysis of physiological amino thiols, Dionex recommends cation-exchange chromatography using the OmniPac® PCX columns.

The AminoPac® PA10 is a strong anion-exchange column used, as part of the AAA-DirectTM system, for high-resolution separations of free amino acids, without derivatization. Amino sugars and carbohydrates are detected
simultaneously with amino acids if they are present in the sample. In addition, separation and detection of a wide range of
sugars, phosphorylated amino acids, and common oxidation products of sulfur-containing amino acids can be accomplished by
this means. AAA-Direct is directly compatible with all four of the commonly used protein hydrolysis procedures, and the results compare favorably
with those obtained by the classical Moore and Stein Ninhydrin procedure.

The AminoPac® PA1 is a pellicular anion-exchange column that offers unique separation characteristics for the separation of amino acids
in a variety of specialized applications. The column is particularly well suited for acidic and acid-labile amino acids and
for amino acid pairs not completely resolved by cation-exchange chromatography.
