The DNAPac® PA100 is a high-resolution, anion-exchange column that provides single-base resolution, is stable under denaturing conditions, is rugged and reliable, and can be readily scaled up. In recent years, a dramatic increase has occurred in the synthesis of nucleic acid oligomers, particularly as PCR primers. As a result, the need for high-speed purification and analysis of nucleic acids has grown. The MicroBeadTM resin technology used in the DNAPac columns provides high-resolution separations to ensure maximum peak purity, and makes linear scale-up of your separation easy. For more complex separations, the polymeric nature of the DNAPac PA100 allows great flexibility in selecting the ideal conditions for your separation, including high pH, high temperature, and chaotropic agents. The DNAPac PA100 is packed with a pellicular anion-exchange resin comprised of a 13-µm-diameter nonporous polymeric substrate to which is bound quaternary amine-functionalized MicroBeads. The rapid mass-transport characteristics of this resin result in higher-resolution oligonucleotide separations than are possible with traditional macroporous resins or reversed-phase columns. Several unique features are incorporated into the packing material to provide unit-base resolution of synthetic oligonucleotides to 60 bases and beyond. Using the DNAPac PA100, synthetic oligonucleotides can be screened for production yield and failure sequences on a routine basis. The DNAPac PA100 can even be operated under denaturing conditions. Either high temperature (up to 90 °C) or high-pH eluents (up to pH 12.5) can be used to eliminate hydrogen bonding, allowing resolution of problem sequences such as self-complementary sequences of poly-G stretches.
