Although a variety of HPLC approaches are proposed for characterizing oligosaccharides released from glycoproteins, most approaches prove inadequate for separating complex mixtures and are limited by their nonspecific nature and low limits of detection. Dionex developed an improved HPLC technique for carbohydrate analysis based on high-performance anion-exchange chromatography coupled with pulsed amperometric detection (HPAE-PAD). This technique provides high-resolution separations with sensitive and specific detection, without requiring derivatization.
The CarboPac PA200 has been developed using a very small nonporous resin particle size to provide very high efficiencies. Attached to the resin bead are extremely small MicroBeadTM particles that have been functionalized to provide the anion-exchange characteristics. As a result of the resin's nonporous nature, dispersion is minimal and column efficiency is extremely high. The capacity of the column in maintained by the high surface area imparted by the MicroBeads. This material is then packed into a 25-cm column body, resulting in the extremely high resolution seen with the CarboPac PA200. Because of the small bead size and long column body, the recommended flow rate is 0.5 mL/min. However, the bed is so stable that you can speed up the separation by increasing the flow rate without damaging the column.
As with the CarboPac PA100, separation of oligosaccharides is based on their fine structural differences, such as the composition and sequence of the oligosaccharides, linkage isomerism, degree of sialylation, and degree of branching. In fact, the CarboPac PA200 was developed to provide the same selectivity as the CarboPac PA100 so that methods developed on the CarboPac PA100 can be transferred to the CarboPac PA200, and thereby provide the same selectivity with increased resolution.
The entire manufacturing process (including resin synthesis, amination, and packing of the columns) is carefully controlled to ensure that every CarboPac PA200 delivers high-resolution separations with reproducible performance. CarboPac PA200 columns are individually tested with two isomers of N-acetylneuraminosyl-D-lactose to ensure column-to-column reproducibility, and each lot is tested with a fetuin digest to ensure lot-to-lot consistency.
