Hydrophobic Interaction Chromatography (HIC) is a technique that has been developed exclusively for the separation of proteins and other large biomolecules. These molecules are separated in a decreasing salt gradient, based on differences in their surface hydrophobicity.  Since proteins are stable in the salt solutions used as the mobile phase, the biological activity of proteins is usually maintained in HIC. In contrast, reversed-phase chromatography uses solvents used for elution that often denature the proteins. Using HIC, the separation mechanism is complementary to those of ion-exchange and gel filtration chromatography. HIC can therefore be used effectively, in combination with these two other techniques, for high-resolution separations of proteins from complex mixtures. The power of the ProPac HIC-10 has been demonstrated in applications ranging from the separation of serum proteins, human skeletal muscle total protein, and snake venom proteins to pancreatin and monoclonal antibodies.

The ProPac HIC-10 stationary phase is based on 5-µm ultrahigh purity spherical silica gel particles, with 300-Å pores.   Multiple points of attachment between the functional layer and the silica substrate are used to ensure enhanced hydrolytic stability in highly aqueous media. In addition, a balanced ratio between the hydrophilic (amide) and hydrophobic (alkyl) groups for protein retention, result in a high efficiency, high capacity, multi-purpose chromatographic HIC media for bioseparations. The unique chemistry of the ProPac HIC-10 column offers exceptional selectivity and stability over the range of pH 2.5 to 7.5, and its design ensures long column life and easy cleanup. The entire manufacturing process is carefully controlled to ensure that every Dionex ProPac HIC-10 column delivers reproducible performance.  ProPac HIC-10 columns are individually tested to ensure column-to-column and lot-to-lot reproducibility.
